HEK293 cells (parental, adherent) have a doubling time of 24-34 hours when grown in DMEM supplemented with 10% FBS at 37 degrees C and 5% CO2. Suspension-adapted variants are faster: HEK293T doubles in 18-24 hours, HEK293F (FreeStyle) in approximately 24 hours in serum-free media, and Expi293F in 24-25 hours at higher densities.
HEK293 is the parental adherent line. HEK293T stably expresses SV40 large T antigen, enabling episomal replication of plasmids with an SV40 origin, making it the standard for lentiviral vector production. HEK293F (FreeStyle) is adapted for serum-free suspension culture in shake flasks and bioreactors for large-scale transient protein expression and AAV production.
Use the formula td = (t2 - t1) x ln(2) / ln(N2 / N1), where N1 and N2 are viable cell counts at times t1 and t2. Both measurements must be during exponential (log) growth phase for an accurate result. The specific growth rate is μ = ln(2) / td.
For adherent HEK293 and 293T, seed at 1-4 x 104 cells/cm2. For suspension HEK293F and Expi293F, seed at 0.2-0.5 x 106 cells/mL. For Expi293F transfection, Thermo Fisher recommends seeding at 0.3 x 106/mL and transfecting when cells reach approximately 3.0 x 106/mL.
Expi293F cells can reach 5-6 x 106 viable cells/mL in Expi293 Expression Medium. Do not exceed 5 x 106/mL during routine passaging. HEK293F (FreeStyle) cells typically reach lower maximum densities of approximately 3 x 106/mL.
