What is the Monod equation?

How do you find mumax and Ks from experimental data?

Measure the specific growth rate mu at several limiting-substrate concentrations S (for example from chemostat steady states or batch exponential phases), then fit the pairs to the Monod equation. You can linearise it (Lineweaver-Burk plots 1/mu vs 1/S; Hanes-Woolf plots S/mu vs S; Eadie-Hofstee plots mu vs mu/S) or, preferably, use nonlinear least-squares regression directly on mu = mumax*S/(Ks+S). Is the Monod equation applicable to batch processes?

Yes, but with a caveat. The Monod equation describes balanced growth and is most rigorously determined at steady state in a chemostat, where mu equals the dilution rate. In a batch culture the substrate is usually far above Ks during most of exponential phase, so mu is effectively constant at mumax and the Monod dependence only becomes visible in the brief deceleration phase as S approaches Ks. Monod parameters fitted from batch data are therefore less reliable than chemostat-derived values, and Ks in particular is hard to estimate from batch alone.

What is the difference between the Monod equation and Michaelis-Menten?

They are mathematically identical in form but describe different things. Michaelis-Menten (v = Vmax*S/(Km+S)) is a mechanistic model of single-enzyme kinetics derived from a reaction mechanism, where Km has a defined kinetic meaning. The Monod equation (mu = mumax*S/(Ks+S)) is an empirical, whole-cell description of population growth; Ks is a lumped, apparent constant that reflects transport and many enzymatic steps, not a single binding affinity. So the algebra is the same but Monod is phenomenological while Michaelis-Menten is mechanistic.

What is the half-saturation constant Ks?

Ks is the substrate concentration at which the specific growth rate equals half of mumax. A low Ks means the organism reaches near-maximal growth even at very low substrate levels (high affinity); a high Ks means it needs a lot of substrate to grow quickly. Typical Ks values for carbon substrates are small, for example roughly 1-10 mg/L glucose for E. coli. Because it is measured where the mu-S curve is steep, Ks is sensitive to data quality at low S.

How do you handle substrate inhibition (Haldane kinetics)?

When high substrate concentrations slow growth (common for toxic substrates such as phenol, methanol or high sugar), the Monod equation is extended to the Haldane (Andrews) model: mu = mumax*S / (Ks + S + S^2/Ki), where Ki is the inhibition constant. Unlike Monod, this gives a peaked curve: mu rises, reaches a maximum at S* = sqrt(Ks*Ki), then declines.
