What is the optimal osmolality range for mammalian cell culture?
Most mammalian cell lines (CHO, HEK293, Vero) grow best at 280–320 mOsm/kg, which matches the osmolality of blood plasma. CHO cells tolerate a controlled increase to roughly 350–450 mOsm/kg during the production phase, where mild hyperosmolal stress can raise specific productivity even as growth rate falls. Below about 260 mOsm/kg cells swell and may lyse; well above 450 mOsm/kg growth typically arrests.
How do feed additions raise osmolality during fed-batch culture?
Concentrated feeds add osmotically active solutes — amino acids, glucose, and salts — so each bolus pushes osmolality upward. The result follows a simple mixing rule: final osmolality = (Osm₁ × V₁ + Osm₂ × V₂) / (V₁ + V₂). Base addition (NaOH or Na₂CO₃) to neutralise lactic acid and the accumulation of lactate and ammonia add further sodium and ions, which is why osmolality tends to climb steadily over a fed-batch run.
How does hyperosmolality affect productivity and growth?
Moderate hyperosmolal conditions (roughly 350–450 mOsm/kg) slow cell division but can increase specific productivity (qp) in CHO and other mammalian cells, so an osmolality shift is a common strategy in monoclonal antibody fed-batch processes. The trade-off is reduced viable cell density and viability, and excessive osmolality can trigger apoptosis and alter glycosylation. The net titre depends on balancing the productivity gain against the loss of biomass.
How is osmolality measured?
The standard laboratory method is freezing-point depression osmometry: dissolved solutes lower the freezing point of water in proportion to the total osmolal concentration (1 Osm/kg depresses the freezing point by about 1.86 °C). The instrument supercools a small sample, induces crystallisation, and reads the equilibrium freezing point to report osmolality directly. Vapour-pressure osmometry is an alternative for very small or volatile samples.
What does mOsm/kg mean, and how does it differ from osmolarity?
Osmolality is reported in milliosmoles of solute per kilogram of solvent (mOsm/kg), while osmolarity is per litre of solution (mOsm/L). For dilute biological media the two are nearly identical. Osmolality is preferred in cell culture and clinical work because it is independent of temperature and pressure and is measured directly by freezing-point depression, whereas osmolarity depends on solution volume, which changes with temperature.

Mild hyperosmolal stress (~320-400 mOsm/kg) develops naturally as concentrated feeds are added in fed-batch, and it typically raises qP while slowing growth and lowering peak viable cell density. Above ~400-450 mOsm/kg, viability and biomass drop sharply, which usually offsets any per-cell productivity gain, so a common strategy is to hold a lower osmolality through the growth phase and allow a controlled increase into the productivity phase.
